Where to get water for testing

Does it make a difference where in the tank I collect the water to test it? It occured to me that since I'm taking water from the top of the tank to test I might not be getting information about lower in the tank, but I've got a filter, so theoretically the water should be all mixed? I've had a quick google, but couldn't find anything about it actually where in the tank to collect the water from. Am I being paranoid? Thanks
 
It’s always easier to take from the top of the tank, but it’s all the same.
 
Temperature is (I think) the only thing of importance that will vary from the top (warmer) to the bottom (cooler). The thermometer should be closer to the top than the bottom, as the cooler lower water is natural, and it is warming the entire tank that is the important aspect and you don't want to be using the lower temperature for that. If that makes sense...
