If the weight is spread out over a cabinet bottom board there should be no problem. Be sure to work on the leveling (carpenter's levels work pretty well)... I've found that some hardware stores these days have peel-and-stick vinyl flat sheets for household baseboards and other purposes and this stuff makes a great quick and easy way to add a swath of height to the bottom of the bottom board. Its usually about 1/8 inch thick and can be stuck to itself to double that. Its best to do wide areas of it of course and don't ever make any correction so tall that it would make things unstable.

It really depends on how well the tiles were put down, Also the size of the tiles matter as well, larger area tiles will be more fragile main reason being the adhesive or sealant underneath if it is point placed or area placed.

As you mentioned you could aleviate the problem by putting a base down for the tank to go on I have heard of people using Marine plie the down side of this is it can look a little odd of course this depends on how much work you put into it and your finishing skills.

Of course this all depends on the size of tank you get?

Yeah it will be fine on tile. My 55gal sits on a tile floor with no problems. Tile makes cleaning up spills so much easier.
