I have 6 yellow labs. I am fairly sure that 2 are male and 4 are female. Yesterday I noticed the larger females mouth appears to be a bit puffier than normal (like she is holding something in there)and kinda distended where the chin is (does this make sense?).

Could it be possible she is holding eggs? How long before I see fry? Will I need to do anything?

It certainly sounds like she's holding.

What you do depends on your end goal, whether you want to keep the fry or not. If you do, there's still no need to do much. After about three weeks or so remove her to a holding tank, where she will spit the fry when she is ready, typically after about a month. Then you mov her back to the main tank and continue to raise the fry as normal. If you do not plan on raising them then just leave it - eventually she'll spit, most of the fry will get eaten, and maybe they'll be a couple of resourceful survivors. Don't expect to raise all the broods these guys produce or you'll quickly need to rent a warehouse for all the tanks you'll need!

If you find she swallows the eggs prematurely don't worry, they often do this on this with their first broods.
