is this the item that connects the hose to the intake strainer and output flower?
 
No, I don't think it is.

lock nuts on a fluval are plastic nuts that thread over the pipe and screw on to the check valve connected directly onto the top section of the canister.

Probably would leak without them imo. You could try using hose clips but DO NOT over tighten them.
 
Just checked my 405 and the nuts are part of the check valve.Thats the part straight above the canister
