just put first dose of ammonia in tank how long should i leave it before i test to see what ammonia is at

I usually give it 20 or 30 minutes to mix well. WD

Do you mean your filter can turn over the volume of the tank 5 times per hour? Not sure what you mean by bpm, what's that?

short answer, just about any decent filter (and if you've bothered to figure out a sump system it is probably more than enough) will mix it quite well in the 20 min or so, probably faster but why not wait if you have the time...
