I have 4 females and 2 males. I recently increased the temp to 78 and they started the mating process. I'm wondering how long i should keep the males in with the females to insure everyone has been for lack of a better expression 'knocked up'? or do some people just keep their males in the same tank and isolate the pregnant female just before she drops?

Well, they should all be "knocked up" in a few days or in one day. YOu could seperate the females from the males right when they are going to drop though.
 
I was just thinking that maybe the females will become stressed if she's pregnant and the males are still harassing her
 
Until the females get close to a drop, I leave all of mine together. When a female is about to drop and I want the fry, I move her to a birth tank where the males and the other females won't all be dining on sushi. Once the drop is done, I will move her back unless she is one of my mollies. Mollies are gentle enough with fry that most of them will survive and the girl can take her time recovering.
