Protozin Whitespot Treatment

The last dose was added on wednesday night and it says "Wait 48 hours before changing treatment" should i change the water and do another course of treatment, as though it was day 1? How much water should i siphon out?

I saw one of my platies flick and rub last night, and im not fully convinced that all of the spots have gone.
