There are actually tests you can buy for this just like nitrite, nitrate, pH, etc. I havent seen them around lately (Canada) but they exist. The Aquarium Pharmaceuticals brand isn't horribly accurate. This is one of those more finnicky tests where you really want to be spending a lot of money for accuracy.

I dont think you can but what makes you ask?

Are your fish gasping at the surface?

As long as you have you filter making a ripple across the water surface this should provide ample oxygen because it increases the surface area.
