If it really was white spot I would complete the course that's recommended by the medicine to kill the WS off and stop the others getting WS.

As for the Clown Loach, if you do want another, I'm told you need a tank with 400+ ltr and they should be in groups of 3's at least

How long should I leave it before restocking some fish....

currently I have 5 neon tetra, 1 red dwarf gourami...and I hope my 4 shrimp are still alive but I've only seen one around since I put the white spot medicine in - but they always hide anyway so I'm not sure.

I would wait a month before adding any new fish.
