When you've conditioned her for two weeks and let her see the male for a couple of days through a clear barrier, she's ready! 
After that though, you know she's ready to be released in with the male when she is showing interest in him, has breeding bars (vertical striping, but this will only show up on dark-bodied females), and is full of eggs.
