The quick answer would be NO. Most people dont unplug their Nutrafin units at night and they were not really designed with that in mind, the Co2 levels obtained with Nutrafin canisters don't usually get high enough for it to be critical. But rather than just saying Yes or No, a more accurate reply would depend on the answer to questions like:
What size of tank you are feeding 2 nutrafin units into ?
> Nutrafin recommend 1 canister for 70Ltrs. I used to run 2 nutrafin units 24/7 into a 120Ltr tank without any problems.
What is the Co2 level in the tank ?
> check your PH & KH and use a Co2 chart to get an idea. I'd advise you to do this anyway, even if you dont have problems. If you are going to inject Co2 into your tank, you may as well know how much you are adding !
Are your fish showing any signs of stress due to Co2 ? (Particularly first thing in the morning)
> plants dont use Co2 at night, so if you are running 24/7 the Co2 levels will be highest just before the lights come on.
If you are in any doubt, there are a couple of things you can do to put your mind at rest.
Obviously you can disconnect the Co2 tube, this is easily done if you create a 'break point' with a non return valve.
You could also run a small air pump at night or you may be able to adjust the filter outflow pipe to provide slightly increased surface movement at night.
