What is the difference between RO and deionized water?

Is it recommended to use 100% DI water when setting up a new planted tank and adding the required minerals etc or shoul I mix it with tapwater?

RO is reverse osmosis water and consists of water that passes throught a membrane filter. With a good filter, >95% of metals, ions, minerals, etc will be removed.

DI is de-ionized and uses an exchange resin which absorbs and/or exchanges ions. The two together, that is, RO/DI filter can bring water up to nearly 100% pure.

I recommend that you repost a separate question in the planted section about the water used in planted tanks.
