Ok so I just ordering some lighting for me 44USG aquarium, I ordered Interpet twin T5 PC 55W with daylight (half white, half blue bulbs....as I like these).

My questions is:

Will I have to get a base layer of plant gravel? or can I just use normal gravel and use root tabs and liquid fert... I will also be using Pressuarized CO2

You can use either tbh, personly I tried to go the cheaper root using playsand with root tabs, i regret it. For the sake of a few extra quid, do it right and make your life easier in the long run :)

Is there anything I can mix into my exisiting substrate, as I have added a thin layer about 1/2 a inch of black sand, does the planted substrate have to be right at the bottom or can is be in the middle?
 
Is that all the sand you have? If not, buy some first layer laterite, that's designed to be mixed with a portion of the substrate. Then you just add the remaining substrate on top :)
